چکیده

To investigate possible reasons for the discrepancy between the abundance of 4-methyl components in free sterols and steryl chlorin esters (SCEs) which is commonly observed in sediments, experiments in which the copepod Calanus helgolandicus grazed on the dino ̄agellates Prorocentrum micans and Alexandrium tamarense were carried out. With P. micans all the algal sterols were esteri®ed as SCEs in the faecal pellets, but there was clear discrimination against the uptake of 4-Me sterols. This discrimination was also evident with A. tamarense. To investigate changes in the faecal pellet SCE and the free sterol distributions during pellet ageing in the P. micans experiment, portions were allowed to stand for up to 29 days in seawater in the dark. Although degradation of SCEs occurred, their esteri®ed sterol distribution remained unchanged. The free sterols were degraded more rapidly than the SCEs and changes in the distribution were observed. This provides further laboratory evidence that sedimentary SCE sterols are more robust markers of phytoplankton communities than the corresponding free sterol distributions. Comparison of the SCE abundance in sterilised and unsterilised pellets indicated that the degradation was a result of microbial activity. # 2000 Elsevier Science Ltd.. All rights reserved.

Shading Influence on the Sterol Balance of Nicotiana tabacum L.

Tobacco plants (Nicotiana tabacum L.) were grown in the field and the apex was removed at the 42-day stage. Shading screens were set up which produced 0, 26, 67, and 90% shade. Plants were grown an additional 25 days before leaves from top, middle, and bottom stalk positions were harvested. Each leaf group was analyzed for free sterol, steryl ester, steryl glycoside, and acylsteryl glycoside. T...

The Saccharomyces cerevisiae YLL012/YEH1, YLR020/YEH2, and TGL1 genes encode a novel family of membrane-anchored lipases that are required for steryl ester hydrolysis.

Sterol homeostasis in eukaryotic cells relies on the reciprocal interconversion of free sterols and steryl esters. The formation of steryl esters is well characterized, but the mechanisms that control steryl ester mobilization upon cellular demand are less well understood. We have identified a family of three lipases of Saccharomyces cerevisiae that are required for efficient steryl ester mobil...

Subcellular distribution of steryl ester biosynthesis in spinach leaves.

Higher steryl ester biosynthetic activities were obtained with Triton X-100-phosphatidylcholine-cholesterol mixed micelles than with Tween 80-phosphatidylcholine-cholesterol mixed micelles when incubated with spinach leaf (Spinacia oleracea L.) acetone powder preparations. The best incorporation of [4-(14)C]cholesterol into [4-(14)C]cholesteryl ester was obtained with a Triton X-100-phosphatidy...

Effects of the Hypocholesteremic Agent Trifluperidol

Trifluperidol (TFP), at a concentration of 100 ,M, inhibited the 24-h growth of Saccharomyces cerevisiae by about 30%. Effects on lipid metabolism were investigated by monitoring the incorporation of [1-14C]sodium acetate into various lipid fractions after 4 and 24 h ofgrowth in the presence of several concentrations of TFP. Although little effect was noted on the amount of free sterols, 24-h i...

Effect of Light on Mevalonic Acid Incorporation into the Phytosterols of Nicotiana tabacum L. Seedlings.

Mevalonic acid-2-(14)C was readily incorporated into the free, esterified, and glycosidic sterol fractions of tobacco (Nicotiana tabacum L. var. Burley 21) seedlings. The time course of mevalonic acid-2-(14)C incorporation was different for the various individual sterols. Campesterol and sitosterol (group I) became radioactive as the free sterol and subsequently as the steryl ester. The reverse...
